SOC Broth is a rich media used primarily in the recovery step of Escherichia coli competent cell transformations. Use of SOC Broth maximizes the transformation efficiency of competent cells.
Dissolve 34.46g in 800-900ml of ddH2O stirring gently with heating until completely solubilized. Adjust pH of the medium to the desired level. Add additional water to bring the solution to 1L. Sterilize with 0.2um membrane filter. Aliquot into sterile containers.
Note: This solution can be autoclaved; extended temperatures could cause the solution to darken, which will not affect product usefulness.
Store powdered media at RT. Opened bottles should be capped tightly and kept in a dark, low humidity environment. Prepared media should be kept refrigerated and used within a short period of time.
