Cleavage of amyloid precursor protein (APP) into 40 or 42 amino acid amyloid beta (ab) peptides is a result of the action of two proteases beta- and gamma-secretase. The beta-site APP cleaving enzyme, or BACE1, is a transmembrane aspartic protease that is localized in the golgi and endosomes. Low levels of BACE1 mRNA are found in most adult tissues with a higher level of expression in pancreas.
Applications
Suitable for use in ELISA, Immunoprecipitation and Neutralization. Other applications not tested.
Recommended Dilutions
ELISA: Use in a Direct ELISA. Immunoprecipitation: 25ug/ml to immunoprecipitate BACE-1 from conditioned media of cell culture spike with recombinant human BACE1 or recombinant mouse BACE1. Western Blot after Immunoprecipitation. Neutralization: The Neutralization Dose (ND50) is typically 10ug/ml. It is measured by the ability of the antibody to neutralize recombinant human BACE1 or recombinant mouse BACE1 cleavage of the fluorogenic peptide substrate Mca-SEVNLDAEFRK(Dnp)RR-NH2 (10uM). Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months after receipt at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Immunogen
Recombinant protein corresponding to Thr22-Tyr460 of human BACE1 expressed in NS0 cells.
Form
Supplied as a lyophilized powder from PBS, pH 7.4, 5% trehalose. Reconstitute with 200ul sterile PBS.

Specificity
