Calnexin (CNX), an abundant ~90kD molecular chaperone, is a unglycosylated resident ER transmembrane protein. In mammalian cells, calnexin, together with calreticulin (CRT), plays a key role in glycoprotein folding and its control within the ER, by interacting with folding intermediates via their monoglucosylated glycans. Calnexin associates with newly synthesized monomeric glycoproteins and only recognizes glycoproteins when they are incompletely folded. Calnexin associates with numerous oligomeric protein complexes within the ER, including beta1 and alpha6 integrins, major histocompatibility class I and class II molecules, the antigen receptors expressed on T and B lymphocytes, human thyroperoxidase (hTPO), and acetylcholine receptor. Recent data also suggest that calnexin might be responsible for the prolonged retention of pro-alpha6 integrin within ER compartment. Furthermore, calnexin has been demonstrated to function as a bonafide molecular chaperone capable of interacting with polypeptide segments of folding glycoproteins.

Applications
Suitable for use in Western Blot, Immunoprecipitation, Immunohistochemistry and Immunocytochemistry. Other applications not tested.
Recommended Dilutions
Western Blot (Colorimetric): 1:1000, detects a band of ~90kD Immunohistochemistry: Paraffin sections Optimal dilutions to be determined by researcher.

Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Synthetic peptide corresponding to the sequence near the N-terminus of canine calnexin.
Form
Supplied as a liquid in PBS, pH 7.2, 0.09% sodium azide, 50% glycerol.
Purity
Purified by Protein A affinity chromatography.
Specificity
Recognizes canine calnexin. Species Crossreactivity: human, monkey, mouse, rat, rabbit, guinea pig, hamster, bovine, C. elegans, sheep, chicken, porcine and Xenopus.
