The catenins (a, b, and gamma) are ubiquitously expressed cytoplasmic proteins that associate with E-cadherin to mediate Ca2+ dependent cell-cell adhesion, a process which is regulated by phosphorylation of protein tyrosine residues. In addition to E-cadherin, b-catenin also associates with several other proteins, including N-cadherin, the tumor suppressor gene product APC (adematous polyposis coli), and a-catenin. The role of the cadherins in several human cancers is currently under investigation. b-catenin appears to play an important role in the Wingless-type (Wnt) frizzled protein receptor pathway associated with cancer of the colon.

ApplicationsSuitable for use in Immunofluorescence, ELISA, Western Blot, Immunoprecipitation and Immunohistochemistry. Other applications not tested.
Recommended DilutionWestern Blot: 2.5-5.0ug/ml. Incubate for 2 hours at room temperature.
Immunohistochemistry (acetone-fixed frozen): Incubate for 1 hour at room temperature.
Immunohistochemistry (FFPE): 10.0-20.0ug/ml. Incubate for 1 hour at room temperature.
Immunoprecipitation: 10.0ug/mg.
Optimal dilutions to be determined by the researcher.
Positive ControlMCF-7, HeLa, A431, or MDCK cells. Human breast carcinoma.
Storage and StabilityMay be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
