Protein kinase CK2 or PKCK2 (formerly known as casein kinase 2) is a serine/threonine kinase, and consists of two constitutively active catalytic (a and a’) and two identical regulatory (b) subunits that form an active tetrameric holozyme. CK2 is involved in an array of cellular responses including cell survival, neoplasia and viral infections, and mediates its survival and anti-apoptotic role in part by protecting cellular proteins from caspase-mediated degradation via their phosphorylation. CK2b (25kD) undergoes phosphorylation at serines 2 and 3 in the Nterminal region and serine 209 in the carboxyl terminus. CK2b serine 209 is phosphorylated by p34cdc2 during mitosis.

Applications
Suitable for use in ELISA and Western Blotting. Other applications not tested.
Recommended Dilution
Western Blotting: 1:1000 (10 mini blots) Optimal dilutions to be determined by the researcher.
Positive Control
HeLa cell lysates or K562 cell lysates.
Storage and Stability
May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Immunogen
Synthetic phosphopeptide corresponding to human CK2b that contains serine 209. Species sequence homology: mouse, rat, chicken, pig and rabbit.
Form
Supplied as a liquid in PBS (without Mg2+ and Ca2+), 1mg/ml BSA (IgG, protease free), pH 7.2, 0.05% sodium azide, 50% glycerol.
Purity
Purified by immunoaffinity chromatography.
Specificity
Recognizes human CK2b when phosphorylated at Ser209. Species sequence homology: Other species CK2b (100%). Does not cross react with CK2a [pS360] (mouse), [pS362], [pS370], or CK2b [pS2].
