Human CD45 is expressed on all cells of hematopoietic origin, except erythrocytes. CD45 is a transmembrane tyrosine phosphatase which can exist in at least nine different isoforms resulting from tissue-specific alternative RNA splicing of exons 4-7 of a single gene coding for the various N-terminal peptide segments. The CD45RO isoform contains none of the alternatively spliced exons and is present on most thymocytes, a subpopulation of resting (memory) T cells within both CD4 and CD8 subsets as well as on monocytes and granulocytes. CD45RO plays an essential role in lymphocyte activation.

Applications
Suitable for use in Flow Cytometry and Immunohistochemistry. Other applications not tested.
Recommended Dilutions
Flow Cytometry: 1:40 (10ug/ml). 80ul of diluted C2400-07C labels 5x10e5 ficoll prepared human peripheral blood mononuclear cells. Immunohistochemistry: Fixed paraffin sections. Optimal dilutions to be determined by the researcher.
Flow Cytometry Performance
1. 5x10e5 ficoll prepared human peripheral blood lymphocytes were washed and pre-incubated 5 minutes with 20ul of 250ug/ml human IgG (to block non specific binding). 2. After which they were incubated 45 minutes on ice with 80ul of C2400-70C at a 1:50 (10ug/ml) dilution. 3. Cells were washed three times, fixed and analyzed by FACS. A net 28% sub population of the cells stained positive with a mean shift of 1.05 log10 fluorescent units when compared to a Mouse IgG2a/FITC negative control at a similar concentration. Binding was blocked when pre-incubated 10 minutes with 20ul of 0.5mg/ml anti-CD45RO antibody (Catalog #C2400-70A).
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. FITC conjugates are sensitive to light.

Supplied as a liquid in PBS, pH 7.5, 0.2% BSA, 0.04% sodium azide, 5% glycerol. Labeled with fluorescein isothiocyanate (FITC).

Purified by Protein A affinity chromatography.
Specificity
Recognizes the CD45RO isoform of about 180kD.
