Human CD152 (CTLA-4) is a cell surface glycoprotein expressed at low levels on activated T-cells. CD152 is a high affinity receptor for the costimulatory molecules CD80 (B7-1) and CD86 (B7-2) and appears to function as a negative regulator of T cell activation.

Applications
Suitable for use in Flow Cytometry, ELISA and Immunohistochemistry. Other applications not tested.
Recommended Dilutions
Flow Cytometry (Blocking): 5ug/ml Optimal dilutions to be determined by the researcher.
Flow Cytometry (Blocking) Protocol
C2548-83C Anti-CD152 antibody blocked the binding of C2548-83V3 CD152 Human, IgG2a Mouse Fc, Fusion Protein (PE) to CD80 and CD86 expressed on human Raji cells. 1. Five x 10e5 cultured Raji human tumor cells were washed and incubated 45 minutes on ice with 80ul of C2548-83V3 at a concentration of 1ug/ml. 2. Cells were washed three times, fixed and analyzed by FACS. Cells stained positive with a mean shift of 1.79 log10 fluorescent units when compared to a Mouse IgG2a Isotype control (PE) at a similar concentration. Binding was 98.6% blocked when C2548-83V3 CD152 Human, IgG2a Mouse Fc, Fusion Protein (PE) was pre incubated 10 minutes with a five fold excess (w/w) of C2548-83C CD152, Blocking (5ug/ml).
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Immunogen
Stimulated human PBL's and human CD152 Ig fusion protein.
Form
Supplied as a liquid in PBS, pH 7.5, 0.5mg/ml gentamicin sulfate.
Purity
Purified by Protein A affinity chromatography from (low FBS containing) tissue culture supernatant. Purity was > 95% immunoglobulin by SDS-PAGE and < 1% bovine Immunoglobulin.
Specificity
Recognizes CTLA-4 expressed on activated T cells. Blocks binding of CD152 Human, IgG2a Mouse Fc, Fusion Protein to its CD80/CD86 receptor.
