Type I collagen is present in soft connective tissue and bone where it represents approximately 90% of the organic matrix. Type I collagen is produced from procollagen when two teloterminal propeptides are digested from the procursor and released into the circulation. Defects in one or both alleles of the type I collagen gene causes “brittle bone” disorder (osteogenesis imperfecta). Patients suffering from such defects have low levels of fully functional type I collagen resulting in bones that break very easily.

ApplicationsSuitable for use in ELISA, Western Blot and Immunohistochemistry. Other applications not tested.
Recommended DilutionsELISA: 1:16,000
Western Blot: Detects human Type I Collagen only in its native triple helix form.
Immunohistochemistry: Used on unfixed, frozen sections of human (1, 4) and bovine (2, 3) skin. If fixation is required, acetone or ethanol is recommended.
Optimal dilutions to be determined by the researcher.

Culture MediumRPMI 1640 with 10% fetal calf serum.
Storage and StabilityMay be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

ImmunogenPepsin soluble, salt-fractionated human placenta type 1 collagen. Epitope: Located near the C-terminal of the molecule.
SpecificityRecognizes native human type I collagen. Does not crossreact with type II, III, V or VI collagens in ELISA or Western Blot. There is no evidence of cross-reactivity with other connnective tissue proteins (laminin, fibronectin, elastin). Species Crossreactivity: Bovine, ovine, porcine, canine and goat type I collagens. Does not react with mouse, guinea pig, feline, rabbit, squirrel, equine, kangaroo or chicken type I collagen.
