Clone Host Grade Applications
Monoclonal Mouse Affinity Purified E B IH FC
Collagens are highly conserved throughout evolution and are characterised by an uninterrupted "Glycine X Y" triplet repeat that is a necessary part of the triple helical structure. Type I collagen (95kD) is found in bone, cornea, skin and tendon. Mutations in the encoding gene are associated with osteogenesis imperfecta, Ehlers Danlos syndrome and idiopathic osteoporosis. Reciprocal translocations between chromosomes 17 and 22, where this gene and the gene for Platelet-derived growth factor beta are located, are associated with a particular type of skin tumor called dermatofibrosarcoma protuberans, resulting from unregulated expression of the growth factor.
Catalog #C7510-17T
ApplicationsSuitable for use in ELISA, Immunohistochemistry, Flow Cytometry and Western Blot. Other applications have not been tested.
Recommended DilutionsELISA (Capture): 2-4 ug/ml
ELISA (Detection): 10ug/ml will detect 50ng of human Collagen Type I with no reactivity with other Collagens. Higher levels of antibody may be necessary to detect lower concentrations of Collagen.
Immunohistochemistry (Unfixed frozen sections): 1:100-1:500
Flow Cytometry: 1:100
Western Blot (Non-denatured and non-reduced): 1:1000
Optimal dilutions to be determined by the researcher.
Positive ControlHuman kidney lysate
Storage and StabilityMay be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

SpecificityRecognizes human Collagen Type I near the C-terminal. Reacts only with native, non-denatured Collegen 1. Displays a high affinity for human, bovine and sheep Type I Collagens. There is no evidence for crossreactivity with Collagen Types III, V and VI or connective tissue proteins (Elastin, Fibronectin and Laminin) at suggested working concentrations. Species Crossreactivity: bovine, sheep, porcine and canine. Does not react with mouse, rat, guinea pig, ground squirrel, equine, kangaroo or chicken.
