| Cyclooxygenase (COX) is a membrane-bound enzyme, which has a molecular weight of 71kD. It is an enzyme that catalyzes the synthesis of cyclic endoperoxides from arachidonic acid to form prostaglandins (lipid-soluble, hormone-like regulatory molecules). Two isoforms of COX have been identified and are known as COX-1 and COX-2. COX-2 is dimeric and is the inducible (i.e., sites of inflammation) isoform of cyclooxygenase . It is normally localized to the endoplasmic reticulum and the nuclear envelope. COX-2 is upregulated in response to endotoxins, pro-inflammatory cytokins, growth factors, hormones, and tumor promoters and in carcinomas, including, sporadic colorectal, breast, pancreatic and cervical carcinomas. It is thought that COX-2 has a role in apoptosis, as well as in carcinogenesis. COX-2 is also constitutively expressed in the brain and kidney.
