| In eukaroytic cells, the cell cycle is mediated by the sequential, activation and inactivation of complexes containing cyclin dependent kinases (CDKs) and cyclins. A number of CDKs have been identified and the activity of these various CDK family members is regulated by multiple mechanisms including binding to specific cyclin proteins, binding to specific small molecular weight inhibitory protein, and reversible phosphorylation. Binding of different cyclin protein is required to activate the catalytically inactive CDKs and determines the substrate specificity of the kinase. On the other hand, CDK inhibitors exert their inhibitory effect by blocking the catalytic activity of the cyclin-bound CDKs. Since the concentration of the CDKs are in excess and remain fairly constant throughout the cell cycle, their activation is determined by the availability of the different cyclin proteins and inhibitory factors. |
