Applications
Suitable for use in Immunohistochemistry (frozen and paraffin-embedded tissue and cytological material) and Immunoblot (Western).
Recommended Dilution
Immunohistochemistry: 1:10; Incubation Time 1h at RT, extended with paraffin sections (overnight at 2-8°C). Pretreatment with paraffin embedded sections, protease pretreatment is required prior to antibody application.
Storage and Stability
Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Immunogen
Human cytokeratin 8, purified from SDS PAGE.
Form
Supplied as a lyophilized powder in PBS, pH 7.4, 0.5% BSA, 0.09% sodium azide. Reconstitute with 1ml sterile ddH2O.
Purity
Purified by Protein A affinity chromatography.
Specificity
Reactivities on Cultured MCF-7, RT 112, HT-29, Detroit 562, RPMI 2650. Cell lines (tested so far) SSC-12, bovine BMGE+H, BMGE-H, MDBK. Represents an excellent marker for distinguishing carcinomas from all non-epithelial tumors. Specifically reacts with cytokeratins 5 and 8 present in nearly all epithelia. Polypeptide reacting: Mr 52,500, Mr 58,000 cytokeratins (type II cytokeratins nos. 5+8) of human epithelial cells. Epitope has been mapped to aa353-367 on alpha helical rod domain. Species Crossreactivity: Human, bovine, murine, amphibia, porcine, hydra.
