The detection of estrogen (ER) and progesterone (PR) receptors using immunohistochemical staining of formalin fixed, paraffin embedded (FFPE) tissue, has gradually replaced ligand binding assays, to become the most common method for the determination of the ER/PR status of breast tumors (Yaziji et al. 2008). ~75% to 80% of breast tumors have estrogen and/or progesterone receptors, and the presence of these receptors helps determine both the patient’s prognosis and the effectiveness of hormonal therapy (Bhargava et al. 2012).

Applications
Suitable for use in Immunoprecipitation, Western Blot and Immunohistochemistry. Other applications not tested.
Recommended Dilutions
Immunohistochemistry (Frozen): 1:40-1:60, the use of Zamboni's fixative is recommended Immunohistochemistry (Paraffin): 1:40-1:80, requires antigen retrieval using heat treatment prior to staining of paraffin sections. It is recommended to use sodium citrate buffer pH 6.0 for this purpose. Western Blot: 1:50-1:100 for 2 hrs. at RT Immunoprecipitation (denatured verified): 10ul/mg protein lysate. Use Protein G. Optimal dilutions to be determined by the researcher.
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
