Hyaluronan and Proteoglycan Link Protein 4, (HAPLN4), also known as brain link protein 2, is one of four secreted HAPLN family proteoglycans that bind hyaluronan. HAPLN4 contains one Ig-type C2-set domain and two link domains. It is predominantly expressed in brain and participates in the development of the perineuronal matrix. Human and mouse HAPLN4 share 91% aa sequence identity.
Applications
Suitable for use in ELISA and Western Blot. Other applications not tested.
Recommended Dilution
Direct ELISA: 0.5-1ug/ml The detection limit is ~5ng/well. Western Blot: 1-2ug/ml Using a colorimetric detection system, the detection kimit is ~1ng/lane and ~25ng/lane under non-reducing and reducing conditions respectively. Chemilunescent detection will increase sensitivity by 5-50 fold. Optimal dilutions to be determined by the researcher.
Hybridoma
The fusion of mouse myeloma with B cells obtained from a mouse immunized with purified, NS0-derived, recombinant human HAPLN4.
Storage and Stability
Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile 40-50% glycerol, PBS. Aliquot and store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Supplied as a lyophilized powder from a 0.2um filtered solution in PBS with 5% trehalose. Reconstitute with 200ul sterile PBS.

Specificity
Recognizes human HAPLN4. Does not react with recombinant human HAPLN1.
