Vitronectin Receptor

The integrin family of cell adhesion receptors consists of a least 16 membrane-associated heterodimers, composed of an alpha and beta subunit that associate in a non-covalent manner. The structure and functional diversity of the integrin family are based on the pairing abilities of the alpha and beta subunits. The integrins are involved in vascular proliferation, adhesion and wound repair. AlphaVbeta3 appears to be selectively expressed on growing blood vessels and has been identified as a marker of angiogenic vascular tissue.

Applications
Suitable for use in Immunoprecipitation, Immunofluorescence, Flow Cytometry, Immunohistochemistry, Immunocytochemistry and Blocking. Not suitable for paraffin embedded tissues. Other applications have not been tested.
Recommended Dilutions
Immunoprecipitation: Precipitates integrin alphaV beta3 complex. When resolved by SDS-PAGE under reducing conditions, bands are observed at ~130kD and ~105kD corresponding to the alphaV and beta3 subunits, respectively. Immunofluorescence: 5-10ug/ml for staining of bFGF-treated 6um cryosections of chick chorioallantoic membrane fixed with acetone. Also effective on 4% paraformaldehyde fixed, frozen tissues and cells. Not effective for immunohistochemical staining of paraffin-embedded tissue sections. Flow Cytometry: 1-3ug/0.5x10e6 cells in 100ul volume Blocking: Inhibits adhesion of cells to vitronectin-coated surfaces at 10-25ug/ml Optimal dilutions to be determined by the researcher.

Human myeloma cell lines M21 and MoalphaV
Recommended Negative Control
Mo cells (this cell line was derived from M21 but does not express integrin alphaV)
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Purified adhesion receptor from the human melanoma cell line M21.
Form
Supplied as a liquid in PBS, pH 7.1. No preservative added.
Purity
Purified by Protein A affinity chromatography.
Specificity
Recognizes the human vitronectin receptor aVb3 complex, an RGD-directed adhesion receptor. Species Crossreactivity: avian, bovine, canine, chicken, monkey, porcine and rabbit. Not reactive with rat.
