Kaiso is a transcriptional represor that belongs to POZ-zinc finger family of transcription factors implicated in development and cancer. It binds to specific methylated CpG dinucleotides in the consensus sequence 5'-CGCG-3' and it also binds to non-methylated consensus sequence 5'-CTGCNA-3'. It recruits the N-CoR repressor complex to promote histone deacetylation and the formation of represive chromatin structures in taget gene promoters. Kaiso may contribute to the repression of target genes of the Wnt signaling pathway.

Kaiso is a novel member of the BTB/POZ (Broad complex, Tramtrak, Bric à brac/Pox virus and zinc finger) zinc finger superfamily of transcription factors. Yeast 2-hybrid studies have shown that Kaiso interacts with catenin p120ctn. This interaction involves the Arm repeats of 1-7 of the p120ctn Armadillo domain and the carboxyl-terminal third of Kaiso.
Applications
Suitable for use in Immunoprecipitation, Immunofluorescence, Western Blot and ELISA. Other applications have not been tested.
Recommended Dilution
Immunoprecipitation: 10ug/IP reaction Immunofluorescence: 10ug/ml Western Blot: 1-3ug/ml ELISA: 0.1-1ug/ml Note: Immunofluorescent assays were performed on cell lines fixed with 100% methanol at –20 °C for 7 minutes. Optimal dilutions to be determined by the researcher.
Storage and Stability
May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Immunogen
Fusion protein encompassing the N-terminus of mouse Kaiso.
Form
Supplied as a liquid in PBS, pH 7.4, containing 0.09% sodium azide
Purity
Purified by Protein A chromatography
Specificity
Reacts with the ~95kD Kaiso protein. Reactivity is confirmed with mouse mammary adenocarcinoma cell line CSML0. Reactivity has been confirmed in MDCK (dog), NBTII (rat), HCT116 (human) and chicken embryo fibroblasts.
