Lat is an adapter protein expressed in lymphoid cells, which is a substrate for ZAP70 and Syk protein kinases. Phosphorylated LAT associates with GRB2, PLC-gamma1 and P85 PI 3-K. Expression of a mutant LAT protein missing tyrosine phosphorylation sites leads to inhibition of T-cell activation, suggesting that the protein is an important signal transducer in T-cells

Applications
Suitable for use in Western Blot and Immunoprecipitation. Other applications have not been tested.
Recommended Dilutions
Western Blot: 2-4ug/ml detects LAT in RIPA lysates from Jurkat cells. Jurkat cell lysate was resolved by electrophoresis, transfered to PVDF and probed with L1390 (2ug/ml). Proteins were visualized using IgG (HRP) goat anti-rabbit secondary antibody and a chemiluminescence detection system. Immunoprecipitation: 8-10ug immunoprecipitated LAT from 500ug of Jurkat RIPA lysate Optimal dilutions to be determined by the researcher.
Included Positive Antigen Control
J9000: Jurkat lysate. Add 2.5ul of 2-mercaptoethanol/100ul of lysate and boil for 5 minutes to reduce the preparation. Load 20ug of reduced lysate per lane for minigels.
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

GST fusion protein corresponding to residues 31-233 of human LAT.
