| Lat is an adapter protein expressed in lymphoid cells, which is a substrate for ZAP70 and Syk protein kinases. Phosphorylated LAT associates with GRB2, PLC-gamma1 and P85 PI 3-K. Expression of a mutant LAT protein missing tyrosine phosphorylation sites leads to inhibition of T-cell activation, suggesting that the protein is an important signal transducer in T-cells | | | Catalog # | L1390 | | Applications | Suitable for use in Western Blot and Immunoprecipitation. Other applications have not been tested. | | Recommended Dilution | Western Blot: 2-4ug/ml detects LAT in RIPA lysates from jurkat cells. Jurkat cell lysate was resolved by electrophoresis, transfered to nitrocellulose and probed with L1390 (0.6ug/ml). Proteins were visualized using goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence. | | Immunoprecipitation: 8-10ug immunoprecipitated LAT from 500ug of Jurkat RIPA lysate | | Optimal dilutions to be determined by the researcher. | | Positive Antigen Control | J9000 Jurkat lysate. | | Storage and Stability | May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. | | CAS Number | n/a | | Clone Type | Polyclonal | | Isotype | IgG | | Host | Rabbit | | Source | Human | | Concentration | ~1mg/ml | | Form | Supplied as a liquid in 0.01M Tris-glycine, pH 7.4, 0.15M sodium chloride, 0.05% sodium azide. before the addition of glycerol to 30%. | | Purity | Purified by Protein A affinity chromatography. | | Immunogen | GST fusion protein corresponding to residues 31-233 of human LAT. | | Specificity | Specific for both forms of human LAT (36 and 38kD). Species Crossreactivity: Mouse. | | | Important Note | This product as supplied is intended for research use only, not for use in human, therapeutic or diagnostic applications without the expressed written authorization of United States Biological. |
