Lat is an adapter protein expressed in lymphoid cells, which is a substrate for ZAP70 and Syk protein kinases. Phosphorylated LAT associates with GRB2, PLC-gamma1 and P85 PI 3-K. Expression of a mutant LAT protein missing tyrosine phosphorylation sites leads to inhibition of T-cell activation, suggesting that the protein is an important signal transducer in T-cells

ApplicationsSuitable for use in Western Blot and Immunoprecipitation. Other applications have not been tested.
Recommended DilutionWestern Blot: 2-4ug/ml detects LAT in RIPA lysates from jurkat cells. Jurkat cell lysate was resolved by electrophoresis, transfered to nitrocellulose and probed with L1390 (0.6ug/ml). Proteins were visualized using goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence.
Immunoprecipitation: 8-10ug immunoprecipitated LAT from 500ug of Jurkat RIPA lysate
Optimal dilutions to be determined by the researcher.
Positive Antigen ControlJ9000 Jurkat lysate.
Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
