Apoptosis, or programmed cell death, is related to many diseases, such as cancer. Apoptosis is triggered by a variety of stimuli including members in the TNF family and prevented by the inhibitor of apoptosis (IAP) proteins. IAP proteins form a conserved gene family that binds to and inhibits cell death proteases. A novel member in the IAP protein family was recently identified and designated Livin and KIAP for kidney IAP (1,2). Livin/XIAP contains a single baculoviral IAP repeat (BIR) domain and a RING finger domain and has two isoforms termed Livin-a and Livin-b (1,3). Transfection of Livin in cells resulted in protection from apoptosis induced by FADD, BAX, RIP, RIP3 and DR6 (1). Livin has direct interaction with several caspases including caspase-3, -7, and Ð9. Livin inhibits the activation of caspase-9 induced by Apaf-1, cytochrome c, and dATP. The two isoforms of Livin appear to have different functions and tissue distributions.

Applications
Suitable for use in ELISA, Immunofluorescence, Immunohistochemistry and Western Blot. Other applications not tested.
Recommended Dilutions
Western Blot: 0.5ug/ml. A band at 33kD (predicted MW) can be detected. A lower but much weaker ban at 30kD was detected in Raji cell lysate, which may represent the short form of Livin. Immunohistochemistry: 5ug/ml. Immunofluorescence: 20ug/ml. Optimal dilutions to be determined by the researcher.

Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Synthetic 17-amino acid peptide near the C-terminus of human Livin.

Supplied as a liquid in PBS containing 0.02% sodium azide.
Purity
Purified by immunoaffinity chromatography.
Specificity
Recognizes human Livin.
