| MMPs are a group of enzymes involved in matrix degradation. They share some important characteristics, such as a common mode of activation, a conserved amino acid sequence in the putative metal binding-active site region, and inhibition by specific proteinase inhibitors known as tissue inhibitors of metalloproteinases (TIMPs). Degradation of the extracellular matrix (ECM) is an essential step in tumor invasion and metastasis. The MMPs each have different substrate specificities within the ECM and are important in its degradation. The enzyme is localized within the cytoplasm. | | | Catalog # | M2425-28 | | Applications | Suitable for use in Immunohistochemistry and Western Blot. Other applications have not been tested. | | Recommended Dilution | Immunohistology (Formalin/paraffin): 10-20ug/ml for 30 minutes at RT No special pretreatment is required for histochemical staining of formalin/paraffin tissues. | | Western Blot: 2-4ug/ml for 2 hours at RT | | Optimal dilutions to be determined by the researcher. | | Positive Control for Western Blot | Conditioned, serum-free medium from TPA-treated human fibrosarcoma (HT-1080) cells. Placenta or breast carcinoma. | | Storage and Stability | May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile 40-50% glycerol, aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. | | CAS Number | n/a | | Clone Type | Polyclonal | | Isotype | IgG | | Host | Rabbit | | Source | Human | | Concentration | ~200ug/ml | | Form | Supplied as a liquid in 10mM PBS, pH 7.4, 0.2% BSA, 0.09% sodium azide. | | Purity | Purified by Protein A affinity chromatography. | | Immunogen | A synthetic peptide corresponding to amino acids 626-644, LFSGRRLWRFDVKAQMVDP of human MMP-9. | | Cellular Localization | Cytoplasm | | Specificity | Recognizes pro (latent) and activated forms of human MMP-9 at 92kD and ~86kD, respectively. Shows no crossreaction with pro and active forms of other MMPs. Species Crossreactivity: guinea pig | | | Important Note | This product as supplied is intended for research use only, not for use in human, therapeutic or diagnostic applications without the expressed written authorization of United States Biological. |
