MMPs are a group of enzymes involved in matrix degradation. They share some important characteristics, such as a common mode of activation, a conserved amino acid sequence in the putative metal binding-active site region, and inhibition by specific proteinase inhibitors known as tissue inhibitors of metalloproteinases (TIMPs). Degradation of the extracellular matrix (ECM) is an essential step in tumor invasion and metastasis. The MMPs each have different substrate specificities within the ECM and are important in its degradation. The enzyme is localized within the cytoplasm.

ApplicationsSuitable for use in Immunohistochemistry and Western Blot. Other applications have not been tested.
Recommended DilutionImmunohistology (Formalin/paraffin): 10-20ug/ml for 30 minutes at RT No special pretreatment is required for histochemical staining of formalin/paraffin tissues.
Western Blot: 2-4ug/ml for 2 hours at RT
Optimal dilutions to be determined by the researcher.
Positive Control for Western BlotConditioned, serum-free medium from TPA-treated human fibrosarcoma (HT-1080) cells. Placenta or breast carcinoma.
Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile 40-50% glycerol, aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
