This gene is a target gene of the transcription factor tumor protein p53. The encoded protein is a nuclear phosphoprotein that binds and inhibits transactivation by tumor protein p53, as part of an autoregulatory negative feedback loop. Overexpression of this gene can result in excessive inactivation of tumor protein p53, diminishing its tumor suppressor function. This protein has E3 ubiquitin ligase activity, which targets tumor protein p53 for proteasomal degradation. This protein also affects the cell cycle, apoptosis, and tumorigenesis through interactions with other proteins, including retinoblastoma 1 and ribosomal protein L5. More than 40 different alternatively spliced transcript variants have been isolated from both tumor and normal tissues.

Applications
Suitable for use in ELISA and Western Blot. Other applications have not been tested.
Recommended Dilutions
Peptide ELISA Titer: 1:64,000 Western Blot: ~55kD band observed in Human Liver lysates (calculated MW of 56.0kD according to NP_002383.2). In transfected HEK293 transiently expressing MDM2 bands of approx. 48kD, 60KDa, 70kD and 90kD are observed. These bands are not observed in the non-transfected HEK293. The 80kD band is endogenously epressed in the negative control. Optimal dilutions to be determined by the researcher.
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Synthetic peptide corresponding to C-DELSGERQRKRHKSD, from the internal region of human MDM2 (NP_002383.2).

Supplied as a liquid in Tris saline, pH 7.2, 0.5% BSA, 0.02% sodium azide.
Purity
Purified by peptide affinity chromatography.
Specificity
Recognizes human MDM2. Species sequence homology: canine.
