MHC Class I molecules are histocompatibility antigens encoded by the H-2 gene complex and consist of heterodimers of highly polymorphic alpha chains noncovalently associated with the invariant beta2-microglobulin. These antigens are expressed on most nucleated cells but expression varies on different cell types. MHC Class I molecules present endogenously synthesized peptides to CD8+ T lymphocytes, which are usually cytotoxic T cells. MHC Class I antigens expressed on thymic epithelial cells regulate the positive and negative selection of CD8+ T cells during T cell ontogeny.

Recommended DilutionFlow Cytometry: 1ug/10e6 cells
Immunohistochemistry: Acetone-fixed, frozen tissue sections
Optimal dilutions to be determined by the researcher.
Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20°C. Aliquots are stable for at least 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

SpecificityRecognizes an epitope in the alpha3 domain that is common to H-2Kd and H-2Dd. Binds to a common determinant in the alpha3 domains of H-2Kd and H-2Dd in the presence or absence of b2 microglobulin. It crossreacts with the alpha3 domain of H-2Kb.
