Myelin basic protein (MBP) is a single-chain, flexible polypeptide of about 18.5kD existing in its natural environment as an extrinsic and loosely bound component of the cytoplasmic portion of the myelin membrane sandwich. Ultrastructural immunocytochemistry with anti-myelin basic protein has shown that MBP is localized in the compact myelin sheath. MBP has not been demonstrated in rough endoplasmic reticulum, lysosomes or any other cytoplasmic organelles. MBP serves as a marker for oligodendrocytes and Schwann cells. MBP is also a marker for malignant schwannomas and appears to play a significant role in the etiology of multiple sclerosis and experimental autoimmune encephalomyelitis.

Applications
Suitable for use in Western Blot, Protein Kinase Assay and Immunocytochemistry. Other applications have not been tested.
Recommended Dilutions
Western Blot: 0.5-2ug/ml. Strongly detects phosphorylated MBP. Note: Some preparations of MBP contain basal levels of phosphorylated MBP, which are detected by the M9758-05. Protein Kinase Assay: Phosphorylated MBP was detected in a direct kinase assay using non-radioactive MAP Kinase Assay Kit, BioAssay™ (M2353) and usign MAP Kinase Immunoprecipitation Kinase Assay Kit, BioAssay™ (M2353-50). Immunocytochemistry: Reported by an independent lab (3). Optimal dilutions to be determined by the researcher.
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Immunogen
Synthetic peptide corresponding to the human myelin basic protein sequence phosphorylated at Thr98 (tuberculin coupled).

Purity
Purified by Protein A affinity chromatography.
Specificity
Recognizes human phosphorylated MBP and most common vertebrate species.
