MPO is an important component of azurophilic granules in neutrophils, being involved in microbicidal processes. The protein is a multimer of 2 heavy chains (55kD) and two light chains (15kD), the heavy chains being linked by a disulfide bond.
Applications
Suitable for use in Flow Cytometry, Immunohistochemistry, ELISA and Western Blot. Other applications not tested.
Recommended Dilution
Flow Cytometry: 1:50-1:100. Membrane permeabilization is required. Immunohistochemistry (Frozen): 1:1000-1:5000 Immunohistochemistry (Paraffin): 1:500-1:1000 Western Blot: Non-reducing conditions. Optimal working dilutions to be determined by researcher.

Hybridoma
Spleen cells from immunized mice were fused with cells of the mouse X63 AG8-653 myeloma cell line.
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Specificity
Recognizes human myeloperoxidase (MPO). Recognizes native MPO in Western Blots and the heavy chain following boiling of the sample. Recognizes recombinant MPO in Western Blots and weakly in ELISA. The antibody may be of value in the study of myeloid cells and myeloid leukemias by Flow Cytometry following cell permeabilization.
