| Myosin is a 480kD protein that interacts with actin in muscle and non muscle cells (8). Conventional myosin contains two identical heavy chains (MHC-200kD each) and four light chains (MLC-15-26kD). Conventional myosin molecules consist of two major regions: tail (rod) and globular heads; they aggregate into filaments through the tail region and interact with actin and with adenosine triphosphate (ATP) through the head region. Multiple forms of myosin heavy chains exist in skeletal, smooth and cardiac muscle and in non-muscle tissues. A spectrum of skeletal muscle fiber types is found in adult skeletal muscles. The two major skeletal muscle fiber types are type I (slow-twitch) and type II (fast-twitch). Skeletal muscle type II fibers can be further subdivided into types IIa (fast red) and IIb (fast white). | | | Catalog # | M9848-05F | | Applications | Suitable for use in Western Blot and Immunohistochemistry (Fr) (paraffin-embedded sections). Other applications not tested. | | Recommended Dilution | Optimal dilutions to be determined by the researcher. | | Storage and Stability | May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot Store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. | | CAS Number | n/a | | Clone Type | Monoclonal | | Isotype | IgG1,k | | Clone No | 8L878 | | Host | Mouse | | Source | Rabbit | | Concentration | As reported | | Form | 20 mM tris-borate, 150 mM sodium chloride | | Purity | Purified | | Immunogen | Rabbit muscle myosin. | | Specificity | Recognizes an epitope in adult rat skeletal myosin heavy chains IIa, IIb and IId (IIx) (in fast-twitch fibers)(1). It also reacts with neonatal but not with embryonal cardiac myosin heavy chains(1). It does not cross react with non-muscle, and smooth muscle myosins. A similar reactivity pattern is observed in human, mouse and chicken skeletal muscle preparations.(2-7). This antibody localizes an epitope on the myosin chain that is stable to the routine formalin fixation. Anti-skeletal myosin to fast twitch skeletal myosin may be used for detecting cross-striated muscle differentiation in tumors.Monoclonal Anti- Myosin (Skeletal, Fast) is useful not only in fiber typing but also in detection of myogenic tumors (10). It enables retrospective as well as prospective fiber typing using formalin-fixed, paraffin-embedded material Species Crossreactivity: Crossreacts with Human, Rabbit, Rat, Mouse, bovine, Cat, Guinea pig and Chicken. | | | Important Note | This product as supplied is intended for research use only, not for use in human, therapeutic or diagnostic applications without the expressed written authorization of United States Biological. |
