Pigment epithelium-derived factor (PEDF) was first identified as a 50kD secreted protein in conditioned medium from cultured fetal human retinal pigment epithelium (RPE) cells. More recently, PEDF was shown to have neurotrophic activity in the retina in vivo. PEDF is also a neurotrophic and neuroprotective factor in other systems, including cultured cerebellar granule cells, primary hippocampal neurons, spinal cord motor neurons and amphibian photoreceptors cultured in the absence of RPE cells. The binding of PEDF to retinoblastoma and cerebellar granule cells may be receptor-mediated. PEDF has been shown to be a potent inhibitor of angiogenesis in endothelial cell cultures and in rat cornea and retina in vivo.

Applications
Suitable for use in Western Blot. Other applications not tested.
Recommended Dilutions
Western Blot: 0.5-2ug/ml detects PEDF in 40ug of RIPA lysates from Y79 cells. 40ug of Y79 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with P3140 (1ug/ml). Proteins were visualized using an IgG (HRP) goat anti-rabbit and a chemiluminescence detection system. Optimal dilutions to be determined by the researcher.

Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Immunogen
His tagged fusion protein corresponding to full length human PEDF (Pigment Epithelial Derived Factor) expressed in baby hamster kidney cells.
Form
Supplied as a liquid in 0.1M Tris-glycine, pH 7.4, 0.15M sodium chloride, 0.05% sodium azide.
Purity
Purified by Protein A affinity chromatography.
Specificity
Recognizes human PEDF at ~50kD. Additional bands were detected at 66 and 76kD, which were competed away by immunogen. These bands may be post-translationally modified forms. Species Crossreactivity: mouse
