| PPARG is a 505aa nuclear protein belonging to the nuclear hormone receptor family and NR1 subfamily, and contains a nuclear receptor DNA-binding domain. It is a receptor that binds to peroxisome proliferators such as hypolipidemic drugs and fatty acids and controls the peroxisomal beta-oxidation pathway of fatty acids. It is thus a key regulator of adipocyte differentiation and glucose homeostasis. It negatively regulates macrophage activation, inhibiting production of monocyte inflammatory cytokines and regulating both native and acquired immune responses. It forms a heterodimer with the retinoic acid receptor RXRA called adipocyte-specific transcription factor (ARF6) and regulates transcription of various genes. Genetic variation in PPARG may influence body mass index (BMI) and abdominal body fat distribution. Defects in PPARG can lead to numerous diseases including type 2 insulin-resistant diabetes, hypertension, colon cancer, familial partial lipodystrophy type 3 (FPLD3), carotid intimal medial thickness 1 (CIMT1) and obesity.
