The Ras/Raf/MEK/ERK signaling pathway conveys mitogenic and differentiation signals from the cell membrane to the nucleus, and controls the proliferation and differentiation of different cell types.1–3 The Raf kinase inhibitor protein (RKIP) interacts with Raf1, MEK, and ERK both in vitro and in vivo,4 interfering with this signaling cascade. RKIP’s mechanism of action has been identified; it inhibits the Raf/MEK/ERK pathway at the Raf-1/MEK interface, where it dissociates the Raf-1/MEK complex and acts as a competitive inhibitor of MEK phosphorylation by Raf.5

Applications
Suitable for use in ELISA, Western Blot. Other applications not tested.
Recommended Dilution
ELISA: 0.1–1.0ug/ml Western Blot: 1–3ug/ml Optimal dilutions to be determined by the researcher.
Storage and Stability
May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Immunogen
Synthetic peptide derived from the N-terminal region of human RKIP (Raf kinase inhibitor protein).
Form
Supplied as a liquid in PBS, pH 7.4, 0.05% sodium azide.
Purity
Purified by immunoaffinity chromatography.
Specificity
Reacts with human and mouse RKIP. In Western Blots, identifies a single 23kD band. Reactivity has been confirmed with human HEK293 cell lysates and mouse brain homogenates. Based on amino acid sequence homology, expected to react with bovine and monkey RKIP.
