SCGF, also named CLEC11a, is a 47 kD, 328 amino acid, secreted glycoprotein member of the C-type lectin superfamily. Produced by CD33+ stem cells and resting chondrocytes, SCGF acts in combination with other factors to promote erythroid, myeloid and endothelial cell production. Over the region used as immunogen, mouse SCGF is 92% and 83% aa identical to the corresponding rat and human protein sequences, respectively.
Applications
Western Blot: This antibody can be used at 0.1-0.2ug/ml with the appropriate secondary reagents to detect mouse SCGF. The detection limit for rmSCGF is approximately 1 ng/lane and 5 ng/lane under non-reducing and reducing conditions, respectively. Direct ELISA: This antibody can be used at 0.5-1.0ug/ml with the appropriate secondary reagents to detect mouse SCGF. The detection limit for rmSCGF is approximately 0.1 ng/well. Optimal dilutions should be determined by each laboratory for each application.
Storage and Stability
Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Immunogen
NS0-derived rmSCGF
Form
Supplied as a lyophilized powder from PBS, 5% trehalose. Reconstitute with 500ul sterile PBS.
Purity
Purified by immunoaffinity chromatography.
Specificity
Selected for its ability to recognize mouse SCGF in direct ELISAs and Western Blots. In these formats, this antibody shows less than 2% crossreactivity with rhSCGF.
