Tenascin is a high molecular weight, multifunctional, extracellular matrix glycoprotein expressed in association with mesenchymal-epithelial interactions during development and in the neovasculature and stroma of undifferentiated tumors (Erickson and Bourdon 1989).It has been described under a variety of names: cytotactin, hexabrachion protein, J1, myotendinous antigen (MI) and glioma mesenchymal extracellular matrix (GMEM).The tenascin molecule is a disulfide-linked hexamer.The expression of tenascin is associated with development and growth, both normal and pathological, whereas the distribution in normal adult tissue is restricted.

Cellular Localization In immunohistological testing of frozen tissue sections of mouse intestine, the antibody stains the core of the villi but not the epithelial cells.
Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20°C. Aliquots are stable for at least 3 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

SpecificityMTn-12 (Aufderheide <i>et al.</i> 1988) reacts specifically with mouse tenascin. In supernatants of cultured mouse fibroblasts, the antibody precipitates two polypeptides of ~210 and ~260kD. It also recognizes these tenascin chains and the 230kD endoglycosidase F-digested fragment in immunoblotting of tissue extracts and fibroblast lysates under reducing conditions (Aufderheide <i>et al.</i> 1988, Weller <i>et al.</i> 1991). In ELISA, it was shown that the antibody reacts specifically against mouse tenascin, but not against fibronectin and collagen type I. Immunoprecipitation data suggests that molecules of the expected size for tenascin-C are detected by MTn-12, and not other tenascins. One can expect several bands in blots or immunoprecipitations but this is because the antibody can detect several splice variants of tenascin C. Nevertheless, the mapping of the epitope and the other data cannot entirely exclude the possibility that it reacts with other tenascins. This would have to be tested directly with purified tenascins. Species Crossreactivity: Crossreacts with Mouse. Not yet tested in other species.
