The Toll-like receptor (TLR) family in mammal comprises a family of transmembrane proteins characterized by multiple copies of leucine rich repeats in the extracellular domain and IL-1 receptor motif in the cytoplasmic domain. Like its counterparts in Drosophila, TLRs signal through adaptor molecules. The TLR family is a phylogenetically conserved mediator of innate immunity that is essential for microbial recognition. Ten human homologs of TLRs (TLR1-10) have been described. By using a BLAST search, Hemmi et al., 2000 have identified and subsequently isolated a cDNA coding for TLR9. Gene knockout experiments suggest that TLR9 acts as a receptor for unmethylated CpG dinucleotides in the bacterial DNA. Human and mouse TLR9 share an overall aa identity of 75.5%. TLR9 is highly expressed in spleen.

Applications
Suitable for use in Immunofluorescence, Flow Cytometry, Western Blot, Immunohistochemistry and Immunocytochemistry. Other applications not tested.
Recommended Dilutions
Flow Cytometry: 0.5-2ug/10e6 cells Western Blot: 2-5ug/ml Immunofluorescence (IC): 1:10-1:500 Immunohistochemistry (Paraffin): 5ug/ml. Staining of formalin-fixed tissues is enhanced by boiling tissue sections in 10mM sodium citrate buffer, pH 6.0 for 10-20 minutes followed by cooling at RT for 20 minutes Optimal dilutions to be determined by the researcher.

Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

KLH-conjugated synthetic peptide corresponding to aa268-300 of human TLR9 isoform A.
