Tumor Necrosis Factor-alpha (TNF-alpha) is a pleiotrophic pro-inflammatory cytokine secreted by various cells including adipocytes, activated monocytes, macrophages, B cells, T cells and fibroblasts. It belongs to the TNF superfamily of ligands and signals through two receptors, TNFR1 and TNFR2. TNF-alpha is cytotoxic to a wide variety of tumor cells and is an essential factor in mediating the immune response against bacterial infections. TNF-alpha also plays a role in the induction of septic shock, autoimmune diseases, rheumatoid arthritis, inflammation and diabetes.

ApplicationsSuitable for use in Flow Cytometry, Western Blot and Immunohistochemistry. Other applications not tested.
Recommended DilutionImmunohistology (frozen): 1:50. 6um tissue sections fixed in acetone for 10 min (4).
Immunohistochemistry (paraffin): 1:50. Tissue sections were fixed in 5% formalin embedded in paraffin and cut into 2um sections. (5)
Flow Cytometry: 1:50. Cells were fixed in PBA containing 0.2% formaldyde (1).
Western Blot: 1:50 detects a band at ~17kD. A reduced sample treatment and 15% SDS-PAGE were used. (4)
Optimal dilutions to be determined by the researcher.
Storage and StabilityMay be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

SpecificityRecognizes human native and recombinant TNF-alpha. Reacts with soluble (17kD) and membrane (26kD) human TNF-alpha. Does not crossreact with TNF-beta, lymphotoxin or receptor bound TNF-alpha. Species Crossreactivity: mouse, guinea pig and rhesus monkey.
