ABTS is a soluble, safe, convenient, and good overall performing substrate for horseradish peroxidase (HRP) based ELISA assays. ABTS develops an intense blue-green color measurable between 405nm and 410nm. It may be used for both kinetic and endpoint reactions.
ABTS (1.46mM) is supplied in citrate buffer, pH 4.0.
In the presence of hydrogen peroxide and HRP, ABTS is oxidized to a radical cation showing absorption maxima at 820nm, 734nm, 650nm, and 405nm. The latter frequency demonstrates a significant molar extinction coefficient and is generally employed for most ABTS assays.
Stopping the reaction with acid does not alter the 405nm spectrum allowing kinetic and endpoint methods to be measured at the same wavelength.
