Highly selective medium for isolation and quantitation of Bacillus cereus from foods. Appearance: Light greenish-beige, homogenous, free-flowing powder Solubility: Yellow to yellow-green, opaque, complete pH (RT ): 7.2 ± 0.2 Cultural Response: Medium was prepared according to label directions with 5% sheep blood and inoculated with the organisms listed. Approximate inoculum: 1000fcu. Cultures were incubated aerobically at 30 ± 2ºC and examined for growth at 18-48 hours. Bacillus cereus (ATCC 13061): Growth: Fair to good Reaction: Blue colonies with lecithin precipitate (halo) Bacillus subtilis (ATCC 9372): Growth: Suppressed to inhibited Reaction: N/A Directions per Liter Suspend 41g of the medium in 950ml of purified water. Heat with frequent agitation and boil for one minute to completely dissolve the medium. Autoclave for 15 minutes at 121ºC (15psi). Cool to 45-50°C. Aseptically add 50ml of a sterile egg yolk suspension and 2ml of a filter sterilized aqueous solution of polymyxin B. Storage and Stability: Store powdered media at RT. Opened bottles should be capped tightly and kept in a dark, low humidity environment. Prepared media should be kept refrigerated and used within a short period of time.
