Belongs to the Bone-growth regulatory factors that are members of the transforming growth factor-beta (TGF-beta) superfamily of proteins. They are synthesized as large precursor molecules which are cleaved by proteolytic enzymes. The active form can consist of a dimer of two identical proteins or a heterodimer of two related bone morphogenetic proteins. Recombinant human Bone Morphogenetic Protein-2 produced in E. coli is a homodimeric, non-glycosylated polypeptide chain containing 115 amino acids. Amino Acid Sequence: The sequence of the first five N-terminal amino acids was determined and was found to be Met-Gln-Ala-Lys-His. Biological Activity: The ED50 as determined by the cytolysis of MC3T3-E1 cells is <50ng/ml corresponding to a specific activity of 2x10e4 IU/mg. Protein Content: Protein quantitation was carried out by two independent methods: 1. UV spectroscopy at 280nm using the absorbency value of 1.4 as the extinction coefficient for a 0.1% (1mg/ml) solution. 2. Analysis by RP-HPLC, using a standard solution of BMP2 as a Reference Standard. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile 100mM acetic acid. Add 0.1% HSA or BSA for long term storage. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. Molecular Weight: 26018D |
Source: E. coli Purity: 95% by RP-HPLC and SDS-PAGE Silver Stain. Chromatographically purified. Form: Supplied as a powder lyophilized from 10mM sodium citrate, pH 3.5. Reconstitute with sterile 100mM acetic acid to 0.1mg/ml. Add 0.1% HSA or BSA for long term storage. This solution can then be diluted into other aqueous solutions.
