Description: c-erbB-2 is a membrane-associated oncoprotein having considerable sequence homology to EGFR. C-erbB-2 is the second member of the EGF receptor tyrosine kinase family of growth factor receptors. Overexpression of c-erbB-2/HER-2 is reportedly a marker of poor prognosis in node-positive and in defined subsets of node-negative breast cancer. Elevated levels of c-erbB-2/HER-2 has also been implicated in response to chemotherapy. Applications: Suitable for ELISA, Immunoprecipitation, Western Blot and Immunohistochemistry. Other applications have not been tested. Recommended Dilution: Immunoprecipitation: 10ug/mg protein lysate (Use Protein A). Western Blot: 5-10ug/ml for 2 hours at RT. Immunohistochemistry (Formalin/paraffin): 2.5-5ug/ml for 30 minutes at RT. Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 minutes followed by cooling at RT for 20 minutes. Optimal dilutions to be determined by the researcher. Positive Control: SKBR-3 cells, T47D cells or breast carcinomas. Control Peptide: C0015-30: c-erbB2, aa1243-1255, Control Peptide (HER2, neu Receptor) Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. |
