Description: Applications: Suitable for use in ELISA and Immunoblot. Other applications not tested. Recommended Dilutions: Immunoblot: 0.5-2.0ug/ml Optimal dilutions to be determined by the researcher. Immunoblot Analysis: 2ug/ml detected phosphorylated CaM kinase II a subunit, Mr ~50kD, in a rat brain microsomal protein preparation. Pretreating blots with lambda phosphatase (400 units/ml), prior to incubation reduced the signal strength. Treatment with lambda phosphatase had no effect on the signal strength using a non-phosphospeci c CaM Kinase II antibody. A rat brain microsomal preparation was resolved by electrophoresis, transferred to nitrocellulose and incubated without lambda phosphatase or incubated with lambda phosphatase before being probed with anti-phospho-CaM Kinase II (2ug/ml). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
