CD34 (Endothelial Cell Marker, Cluster Designation 34, Hematopoietic Progenitor Cell Antigen CD34, HPCA1)
CD34 is a single-pass type I membrane glycophosphoprotein present on small vessel endothelial cells and hematopoietic progenitor cells. The apparent molecular mass of CD34 is heterogeneous, depending on the glycosylation state in different cell types. In cultured endothelioma cell lysate, CD34 has a MW of ~100kD, whereas in lung lysates it is ~80kD. Two Isoforms of CD34 exist and both are expressed on the cell surface. CD34 is an adhesion molecule performing a role in early hematopoiesis by mediating the attachment of stem cells to the bone marrow extracellular matrix or directly to stromal cells. CD34 acts as a scaffold for the attachment of lineage specific glycans, allowing stem cells to bind to lectins expressed by stromal cells or other marrow components. CD34 presents carbohydrate ligands to selectins. CD34 is widely used as a marker to select early hematopoietic stem and progenitor cells in experimental and clinical hematopoiesis.

Suitable for use in ELISA, Immunofluorescence, Immunohistochemistry, Western Blot, Immunoprecipitation, Flow Cytometry and Cell Separation. Other applications have not been tested.

Recommended Dilution:
Immunohistochemistry (Frozen): 1:50
Immunohistochemistry (Paraffin): 5ug/ml; Boil twice for 5 minutes in citrate, pH 6.0, or 0.03% trypsin treatment for 10 minutes at 37C. Incubation at 4C overnight resulted in the specific staining of endothelial cells.
Western Blot: 1:50; Both reduced and non-reduced, 70-105kD, depending on glycosylation state and cell type.
Immunoprecipitation: 1:50; 100kD protein observed in H5V cells.
Flow Cytometry: 0.1ug per 10e6 cells (mice blood cells).
Optimal dilutions to be determined by the researcher.

Storage and Stability:
May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Recognizes mouse CD34. Recognizes a neuraminidase sensitive epitope on endothelium in vivo, particularly on small vessels and neoformed capillaries and developing vascular structures in embryonal structures. Antibody can be used for identification and characterization of capillary endothelial cells. Reacts with hematopoietic progenitors particularly of myelomonocytic colony forming cells.
