Description: CD45 or Leukocyte Common Antigen (LCA/T200) family is comprised of at least four isoforms (CD45RA, CD45RB, CD45RC, and CD45RO) of membrane glycoproteins with MW of 180-220kD. It consists of a large, heavily glycosylated, amino-terminal extracellular domain of 400-550 amino acids, a single transmembrane region and a large carboxy-terminal cytoplasmic domain of 700 amino acids. CD45/LCA is expressed on hematopoietic cell lines but absent on non-hematopoietic cell lines, normal and malignant non-hematopoietic tissues. The intracellular portion of CD45 proteins has phosphatase activity and is involved in regulation of transmembrane signals, functioning as a leukocyte membrane receptor capable of transducing signals via its phosphotyrosine phosphatase domains. Applications: Suitable for use in Flow Cytometry, Immunohistochemistry and Immunoprecipitation. Other applications not tested. Recommended Dilution: Flow Cytometry: 1:10-1:50; 10ul labels 10e6 cells in 100ul Immunohistochemistry (Frozen): 1:500-1:1000 Immunohistochemistry (Paraffin): 1:100-1:200. Requires antigen retrieval using heat treatment prior to staining of paraffin sections. 1mM EDTA, pH8.0 is recommmeded. Optimal dilutions to be determined by the researcher. Hybridoma: NS1 myeloma cells with spleen cells from Balb/c mice. Recommended Positive Control: Human Tonsil Recommended Negative Control: I1904-83P1: IgG2a Murine Negative Control Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. |
