C-Jun is a component of the transcription factor AP-1 that binds and activates transcription at TRE/AP-1 elements (Figure 1). The transcriptional activity of c-Jun is regulated by phosphorylation at Ser63 and Ser73 (1,2). Extracellular signals including growth factors, transforming oncoproteins, and UV irradiation stimulate phosphorylation of c-Jun at Ser63/73 and activate c-Jun dependent transcription. Mutation of Ser63/73 renders c-Jun nonresponsive to mitogenic and stress induced signaling pathways. The MAP kinase homologue, SAPK/JNK, binds to the N-terminal region of c-Jun and phosphorylates c-Jun at Ser63/73 (3, 4). The c-Jun (Ser63) II and c-Jun (Ser73) Antibody Kit provides reagents and protocols for the rapid analysis of c-Jun phosphorylation at Ser63 and at Ser73. The kit includes two phospho-antibodies that recognize c-Jun only when phosphorylated at Ser63 or Ser73, a c-Jun antibody (phosphorylationstate independent), protein controls and protein markers for western blots, and our HRP Western Detection System.

Suitable for use in Western Blotting, Immunocytochemistry. Other applications not tested.

Recommended Dilution:
Western blotting: 1:1000
Immunocytochemistry: 1:1000

Optimal dilutions to be determined by the researcher.

Storage and Stability:
May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Purity: Purified by protein A and peptide affinity chromatography.
Form: Supplied as a liquid in 10mM sodium HEPES, pH 7.5, 150mM sodium chloride, BSA, 50% glycerol.
Specificity: Detects phosphorylated Ser63 of c-Jun but does not appreciably crossreact with the corresponding phosphorylated form of JunD or JunB. Phospho-c-Jun (Ser73) antibody detects phosphorylated c-Jun at Ser73 or JunD at Ser100, yet will not react with up to 0.5ug of non-phosphorylated c-Jun or JunD.

Species crossreactivity: human, mouse.
