Description: Connexin 40 (Cx40) is a gap junction protein highly expressed by vascular endothelial cells and in the mammalian heart, specifically in the atrial myocardium and the His-Purkinje conduction system. At least three other connexins are expressed in the heart, including Cx37 (endocardium), Cx43 (atrial and ventricular myocardium, distal parts of the conduction system), and Cx45 (throughout the heart). Mice lacking the gene for Cx40 exhibit reduced atrial but normal ventricular conduction velocity; these mice display right bundle-branch block and impaired left bundle-branch conduction. Cx40 and Cx43 frequently co-localize in cells of the cardiovascular system, and participate in the formation of heterotypic gap junction channels. For example, atrial myocytes express both Cx40 and Cx43, which oligomerize into the same hemichannel. Atrial connexons consist of hexomeric connexin assemblies that contain varying proportions of Cx40 and Cx43 proteins. Applications: Suitable for use in Western Blot, Immunofluorescence, and Immunohistochemistry. Other applications have not been tested. Recommended Dilutions: Western Blot: 1-3ug/ml Immunofluorescence: 1-3ug/ml Immunohistochemistry: 1-3ug/ml Note: For IHC mice were perfused transcardially with 3ml pre-fixative (50mM phosphate buffer, 0.1% sodium nitrite, 1u/ml heparin), followed by 40ml fixative (0.16M sodium phosphate buffer, 0.2% picric acid, and 1-,2-, or 4% paraformaldehyde), and perfusion with 10ml sucrose wash. Tissues were then stored in cryoprotectant (25mM PBS, pH 7.4, 0.04% sodium azide, 10% sucrose) for a minimum of 24 hours prior to sectioning. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. |
