Description: The human Dickkopf (Dkk)-related proteins are a structurally and functionally heterogeneous family of proteins. This family is comprised of hDkk1, -2, -3, -4 and Soggy (Sgy), a Dkk-3 related protein. Members of the Dkk-related family display unique patterns of mRNA expression in human and mouse tissues and are secreted when expressed in 293T cells. Several secreted antagonists, including Dickkopf-1 (Dkk-1) (German for “big head, stubborn”) regulate the activity of Wnt proteins secreted glycoproteins that influence many aspects of embryonic development in metazoans. Dkk-1 is a head inducer secreted from the vertebrate head organizer, that induces anterior development of Xenopus by antagonizing Wnt signaling. Dkk-1 inhibits Wnt-induced axis duplication, acts upstream of disheveled3 and antagonizes Wnt signaling upstream of its effect on beta-catenin regulation. Dkk-1 is an LRP6 ligand5 that inhibits Wnt signaling by blocking Wnt-induced Fz-LRP6 complex formation. Dkk-1 plays a crucial role in the control of programmed cell death during vertebrate limb development. Bmp-4 is one of several upstream regulators of Dkk-1 expression. Furthermore, the proapoptotic hDkk-1 gene may play important roles in linking the oncogenic Wnt and p53 tumor suppressor pathways. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 0.1-2ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
