Description: The Dopamine D2 Receptor is one of the five types (D1 to D5) of receptors for dopamine. The activity of this receptor is mediated by G proteins which inhibit adenylyl cyclase. 2 isoforms are reported for the D2 Dopamine Receptor, Isoform 1 and isoform 2 which are expressed in the pituitary and brain; Isoform 1 is expressed seven times more than isoform 2 in the caudate nucleus. Applications: Suitable for use in ELISA, Western Blot, Immunohistochemistry and Immunocytochemistry. Other applications not tested. Recommended Dilutions: Western Blot: 40ug/ml using whole rat brain homogenate resulted in a single band being detected at ~48kD. Immunohistochemistry/Immunocytochemistry: 80ug/ml stains specific cells in various regions of PLP fixed rat brain sections including the Medial septum, Nuclcus accumbens, Dentate gyrus, Globus pallidus, Medial forebrain bundle, Cortex regions 1-3, Substantia Nigra teticulata and the Ventral regmental area. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS, 10mg/ml BSA. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
