Edinburgh Minimal Media is a thiamine-free formulation for nmt1 promoter studies for Schizosaccharomyces pombe. Phosphate Free formulation is used for 32P-phosphate labeling. Sodium phosphate is removed and replaced with potassium hydrogen phthalate with 2g/L sodium acetate (14.6mM). The medium is adjusted to pH 5.5. Directions per Liter: Dissolve 29.328 grams per liter of ddH2O. Heat with stirring until completely solubilized. Adjust pH as necessary. Dispense into appropriate containers. Loosen caps or cover with foil. Autoclave for 15 minutes at 121ºC (15psi). Note: May also sterilize with 0.2um membrane filter. Aliquot into sterile containers. Contains heat-labile compounds that can be damaged with autoclaving. Storage and Stability: Store powdered media at RT. Opened bottles should be capped tightly and kept in a dark, low humidity environment. Prepared media should be kept at 4°C and used within a short period of time.
