Description: HuR (Hu-antigen R, ELAV1) is an RNA binding protein that contains 3 RNA-binding domains. It is known to bind AU-rich elements in mRNA. It regulates gene expression by destabilizing their mRNA. It is known to be upregulated in certain cancers such as breast and gastric cancers. Some of its targets include cFos, IL-3, and BRCA1 mRNAs. HuR has also been shown to be involved in the regulation of VEFG-A and NFkB expression in certain cell types. It activation and accumulation are also linked to many energy signaling molcules such as that of the PI3K/Akt pathway and AMPK. Applications: Suitable for use in Immunohistochemistry, Immunocytochemistry, Western Blot and Immunoprecipitation. Other applications not tested. Recommended Dilutions: Immunohistochemistry: Reported to detect HuR in rabbit placental tissue. Immunocytochemistry: 1:500. Detects HuR in HeLa and NIH/3T3 cell lysates. Western Blot: 1ug/ml. Detected HuR in RIPA lysates from 3T3/A31 cells. 3T3 cell lysate was resolved by electrophoresis, transferred to PVDF and probed with H7953-34. Proteins were visualized using an IgG (HRP) donkey anti-rabbit and a chemiluminescence detection system. Immunoprecipitation: 10ug immunoprecipitated HuR in 3T3/A31 RIPA lysates. Optimal dilutions to be determined by the researcher. Positive Antigen Control (Included): F4209: Fibroblast Cell, 3T3/A31: Add 2.5ul of 2-mercaptoethanol/100ul of lysate and boil for 5 minutes to reduce the preparation. Load 20ug of reduced lysate per lane for minigels. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. |
