Description: Neurofilaments are the intermediate filaments found in neuronal cells. They are composed of a combination of subunit proteins which typically includes the neurofilment triplet proteins, NF-L, NF-M, and NF-H. Neurofilaments may also include peripherin, alpha-internexin (NF66), nestin, and vimentin. Neurofilament proteins are useful biomarkers as their expression can vary during neuronal development and differentiation, between between neuronal cell subtypes, and be altered during pathological states. Alpha-internexin/NF66, related to the neurofilament triplet proteins, is expressed during early neuronal development. The temporal and spatial expression pattern of alpha-internexin varies between different subtypes of differentiating and mature neurons. An upregulation of alpha-internexin has been observed during neuronal regeneration. Abnormal neuronal aggregates of alpha-internexin along with N-L, NF-M, and NF-H have been identified as the pathological hallmarks of the neuronal intermediate filament inclusion disease (NIFID), an early onset neurodegenerative disease associated with dementia. Antibodies to alpha-internexin are useful probes for studying and classifying neuronal subtypes, and for identifying neuronal progenitor cells and other cells committed to the neuronal lineage. Applications: Suitable for use in Immunofluorescence, Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Immunofluorescence: 1:250 Western Blot: 1:5000 Immunohistochemistry: 1:2500 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
