Description: Applications: Suitable for use in Immunohistochemistry, Western Blot and ELISA. Other applications not tested. Recommended Dilution: Immunohistochemistry (fresh frozen): Tissues were sectioned (6um), dried for 1 hour at RT and fixed in 1:1 solution of methanol and acetone. Sections were air dried for 1 hour. Sections may be stored wrapped in aluminium foil at -70°C until used. Immunohistochemistry (Paraffin): 4% PFA (3hr) should be used for skin biopsies. 5um sections are dewaxed and rehydrated with water. A proteinase treatment of 0.1% trypsin for 15 minutes in water should be performed prior to blocking and antibody staining. Note: The antibody reacts better in frozen tissues than in paraffin embedded tissues. Western Blot: Purified normal human eosinophils were treated with digitonin (5mg/ml) for 10 minutes before boiling in sample buffer. Ran on a 15% SDS-PAGE. Purified MBP migrates at ~14kD, with typical human samples ranging from 10-16kD. Optimal dilutions to be determined by the researcher. Source: Tissue culture supernatant Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
