Used as a medium for cultivation of Mycobacterium sp. when supplemented with glycerol and OADC. Modification of Middlebrook 7H10 or 7H9 formulations. Directions per Liter: Mix 19g of medium into 900ml of distilled/deionized (DDI) water and 5ml glycerol. Heat to boiling until dissolved completely. Autoclave for 10 minutes at 121ºC (15psi). Cool to 45-50°C and aseptically add 100ml of OADC enrichment. Storage and Stability: Store powdered media at RT or 4°C. Opened bottles should be capped tightly and kept in a dark, low humidity environment. Prepared media should be kept at 4°C and used within a short period of time. Appearance: Light blue-beige, homogeneous, free- flowing powder Solubility: Grey-white to pale light green, trace to slightly hazy pH (25°C): 6.6 ± 0.2 Expected Culture Response: 3-28 days at 35°C under CO2.
