Description: E. coli-derived MutS is a protein component of the mismatch repair pathway, selectively binding DNA fragments containing mispaired or unpaired bases. MutS binds mismatches that most resemble base pairs (i.e., G:T mismatches in G:C rich regions), allowing MutS to be used for mutation/SNP detection and isolation applications and mismatch repair structure/function studies. Nuclease Activity: No detectable nuclease activity when incubated with single or double stranded DNA for 16 hours at 37°C. Proteinase Activity: No significant degradation of protein as judged by SDS-PAGE analysis, or loss of activity, after 16 hours at 37°C. Applications: Suitable for use in ELISA. Other applications not tested. Recommended Dilutions: ELISA: 1:1000-1:10,000; Recommended dilution buffer for use: 1X PBS, 3% (w/v) BSA (IgG-free BSA). The antibody is unlabeled, therefore detection may be accomplished with any appropriately labeled anti-rabbit secondary antibody. Alternatively, plates may be coated with a 1:1000 dilution of antibody in 0.05M Na2CO3 Buffer, pH 9.6. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. |
