Myelin Basic Protein (MBP)
Myelin basic protein (MBP) is a single-chain, flexible polypeptide of about 18.5kD existing in its natural environment as an extrinsic and loosely bound component of the cytoplasmic portion of the myelin membrane sandwich. Ultrastructural immunocytochemistry with anti-myelin basic protein has shown that MBP is localized in the compact myelin sheath. MBP has not been demonstrated in rough endoplasmic reticulum, lysosomes or any other cytoplasmic organelles. MBP serves as a marker for oligodendrocytes and Schwann cells. MBP is also a marker for malignant schwannomas and appears to play a significant role in the etiology of multiple sclerosis and experimental autoimmune encephalomyelitis.

Suitable for use in Western Blot, MAP Kinase Assay and ELISA. Other applications not tested.

Recommended Dilution:
Western Blot: 0.05-0.5ug/ml detects MBP in RIPA lysates from mouse and rat brain tissue preparations. Mouse brain cytosol homogenate was resolved by electrophoresis, transferred to PVDF and probed with M9758-02F. Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system.
MAP Kinase and Indirect ELISA: 0-200ng of M2362-06 was used to phosphorylate well-bound MBP. Phosphorylation of MBP was detected using M9758-05. Total MBP was detected by M9758-02F.
Optimal dilutions to be determined by the researcher.

Storage and Stability:
May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
