Description: NLGN3 is a 110-114kD member of the type-B carboxyesterase/lipase family of proteins. It is a neuronal transmembrane protein that forms Ca++-dependent intercellular junctions with short b-neurexin isoforms. This seems to contribute to both glutamatergic and GABAergic synapse formation. Mutations in NLGN3 are associated with a reduction in protein expression and the occurrence of autism. Mature human NLGN3 is an 811aa type I transmembrane protein. It contains a 672aa extracellular domain (ECD) aa38-709, plus a 118aa cytoplasmic region. The ECD possesses a nonfunctional carboxyesterase domain aa41-625 Multiple splice variants exist. There is a deletion of aa153-172 that may also be accompanied by an alternative start site at Met118, and a deletion of aa153-192 that may also be accompanied by a five Lys substitution for aa554-848. Over aa39-709, human NLGN3 shares 99aa identity with mouse NLGN3. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 2ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
